药代性质 (ADME/PK)
吸收、分布和排泄
膳食锌约有20%至30%被吸收,主要在十二指肠和回肠吸收。吸收量取决于食物中锌的生物利用度。红肉和牡蛎中的锌生物利用度最高。植酸盐可能通过螯合作用和在碱性pH值下形成不溶性复合物来抑制锌的吸收。吸收后,锌在肠道中与金属硫蛋白结合。内源性锌可在回肠和结肠中被重吸收,形成肠胰循环。锌主要经粪便排出(约占90%);少量经尿液和汗液排出。吸收后,锌在肠道中与金属硫蛋白结合。锌广泛分布于全身。它主要储存在红细胞、白细胞、肌肉、骨骼、皮肤、肾脏、肝脏、胰腺、视网膜和前列腺中。本研究在兔体内比较了硫酸锌和一种新型泛酸锌的药代动力学。两种盐均以3.3 μCi锌-65/kg体重的剂量给予兔子。两种化合物的药代动力学符合双室开放模型。两种盐的尿排泄情况相似,在皮肤和毛发中的分布也相似,但泛酸锌在肝脏中的保留程度低于硫酸锌(ZnSO4)。吸收率:20%~30%。蛋白结合率:99%。排泄途径:经小肠排泄。在妊娠不同阶段的妇女中,每日口服200 mg硫酸锌可使血清锌水平从109.7 mg/dL升高至205.4 mg/dL。对照组血清锌水平从妊娠早期的 113.0 mg/dL 下降至妊娠晚期的 83.8 mg/dL。……据报道,当 10 名健康青年男性一次性服用 45 mg 硫酸锌(Zn2+,以明胶胶囊形式)时,其吸收半衰期为 0.4 小时。在总共 8 小时的研究期间,频繁测量血清浓度。2.3 小时后(tmax)发现血清中 Zn2+ 的平均最大浓度为 8.2 μmol/L。有证据表明存在肠道再循环,在达到 tmax 之前,吸收阶段 1.4 小时后出现第一次反弹效应,平均再吸收率为给药剂量的 70%。随后的反弹效应(最多 5 次)以 1.2 小时的规律间隔出现,且再吸收量逐渐减少。有关硫酸锌(共 14 项)的更多吸收、分布和排泄(完整)数据,请访问 HSDB 记录页面。

药物适应症

作用机制
体外大鼠回肠研究表明,锌通过抑制基底外侧钾(K)通道来抑制cAMP诱导的氯依赖性液体分泌。该研究还表明锌对cAMP激活的K通道具有特异性,因为锌不会阻断钙(Ca)介导的K通道。由于该研究并非在缺锌动物中进行,因此其结果表明,锌在非缺锌状态下可能有效。此外,锌还能改善水和电解质的吸收,促进肠上皮再生,提高刷状缘酶的水平,并增强免疫反应,从而更好地清除病原体。
